John Hamilton Mackenzie (1806-1875) was a lifelong friend of Edgar Allan Poe. He was also the foster brother (and, after his father William Mackenzie's death in 1829, the legal guardian) of Poe's sister, Rosalie Mackenzie Poe. After Poe's death, John H. Mackenzie provided information about the author to the biographer Susan Archer Talley Weiss for her book The Home Life of Poe.

The Poe Museum also owns an albumen print photograph of John Hamilton Mackenzie taken in later years.
